摘要
用HBsAg制备了一株分泌乙型肝炎病毒前S2抗原单克隆抗体(MAb)的杂交瘤细胞株(1E2),小鼠腹水中MAb效价达10^7。1E2MAb有很强的抑制多聚人血清白蛋白(pHSA)和HBsAg中前S2抗原结合的能力;与重组痘苗病毒表达的中分子蛋白(含S和前S2抗原)而不与小分子蛋白(含S抗原)反应;与人工合成的前S2肽段(N端120-146氨基酸序列)特异性结合。
A hybrid cell line producing MAb, which could recognize an epitope encoded by the Pre-S(2) region of HBV genome was obtained by hybridoma technique. The MAb-1E2MAb reacted with Pre-S(2) epitope on middle protein of the viral envelope The specificity of 1E2MAb waa confirmed by its capability of binding to the synthetic pep-tide corresponding to N-terminal 120-146 ammo acid sequence of Pre-S(2) region. lE2MAb was used to develop a sensitive enzymoimmunoassay for determination of Pre-3(2) im the sera of HBV infectious patients. The results showed that the titers of Pre-S(2) and HBsAg in HBsAg positive sera were well correlated (r = 0.87, P< 0.01), especially in HBeAg positive cases (r = 0.94, P<0.01).
