目的检测表皮生长因子受体(EGFR)及黏蛋白MUC5AC在肺气肿大鼠肺组织中的表达,了解EGFR与黏蛋白合成的关系。方法用免疫组织化学染色、Westernblot及RTPCR方法研究吸烟诱导的肺气肿大鼠20例、正常大鼠20例的肺组织EGFR、MUC5AC蛋白和mRNA表达。结果肺气肿组大鼠EGFR蛋白定量和mRNA表达分别为439±084和041±012,均显著高于正常组(分别为250±061和031±008)(P<0001,P<001)。MUC5AC与EGFR表达呈正相关(r=08156,P<001)。结论EGFR参与了吸烟诱导的肺气肿大鼠气道黏蛋白的合成。
Objective To investigate the expression of epidermal growth factor receptor(EGFR) and mucin MUC5AC in the airways of rats with emphysema.Methods EGFR and MUC5AC mRNA expression at both gene and protein levels were examined using immunohistochemical assay,Western blot and RT-PCR in pulmonary specimen in 20 rats with emphysema induced by cigarette smoking and in 20 normal rats.Results A strong EGFR protein signal and mRNA expression(4.39±0.84,0.41±0.12 respectively) were detected in rats with emphysema in comparison with a weak signal in normal animals(2.50±0.61,P<0.001;0.31±0.08,~P< 0.01,respectively).A significant positive correlation was revealed between EGFR mRNA expression and MUC5AC mRNA expression (r=~0.815 6 ,P<0.01).Conclusion These findings suggest a role of EGFR in mucin expression in epithelium of emphysematous rat airway induced by cigarette smoking.
