摘要
目的 :研究不同浓度rhbFGF、rhBMP 2独立或联合对体外培养兔成骨样细胞增殖的影响 ,探讨两者协同作用的可能最适浓度。方法 :取经矿化诱导第三代兔骨髓基质细胞获得的兔成骨样细胞以 2× 10 6/ml的浓度接种于 96孔板 ,用 3种浓度rhFGF、单一rhBMP 2、不同浓度rhFGF和rhBMP 2的组合等 7种条件培养基进行诱导培养 ,设空白对照组 ,每组 8孔。于 1、4、7、10d后用MTT比色法检测细胞增殖情况。结果 :第 4d ,10ngrhbFGF + 10 0ngrhBMP 2 /ml组的MTTOD值与 10ng/mlrhbFGF组无显著性差异 (P >0 .0 5 ) ,但大于 10 0ng/mlrhBMP 2组、1ngrhbFGF + 10 0ngrhBMP 2 /ml组和 5 0ngrhbFGF + 10 0ngrhBMP 2 /ml组 ,且有显著性差异 (P <0 .0 5 )。结论 :rhbFGF和rhBMP 2的不同浓度组合对兔成骨样细胞的增殖有不同影响 ,其中 10ngrhbFGF + 10 0ngrhBMP 2 /ml组合对细胞的增殖有最强的促进作用。
Objective:To study the effect of recombinant human basic fibroblast growth factor(rhbFGF) and recombinant human bone morphogenetic protein-2(rhBMP-2 )with different concentrations on the proliferation of osteoblast-like cells obtained from rabbit bone marrow stromal cells by induced mineralization.Method:The osteoblast-like cells were obtained from rabbit bone marrow stromal cells by induced mineralization. The osteoblast-like cells subcultured to the 2nd passage were used for these experiments. The cells were seeded separately into 96 well plates with approximately 2×10 5 cells/well and incubated in different mediums. The samples were devided into 8 groups by different conditional mediums, included ① Group A:1 ng/ml rhbFGF, ② Group B:10 ng/ml rhbFGF, ③ Group C: 50 ng/ml rhbFGF, ④ Group D: 100 ng/ml rhBMP-2, ⑤ Group E: 1 ng rhbFGF+100ng rhBMP-2/ml, ⑥ Group F: 10 ng rhbFGF+100 ng rhBMP-2/ml, ⑦ Group G: 50 ng rhbFGF+100 ng rhBMP-2/ml, ⑧ Group H: the blank group. Then, the proliferation of osteoblast-like cells were measured after 1day, 4 days, 7days, 10days by MTT methods.Result:The effect was different of the groups of combined growth factors.The effect was the strongest for the proliferation of rabbit osteoblast-like cells with 10ng rhbFGF and 100 ng rhBMP-2/ml.With lower concentration of rhbFGF(1 ng/ml),it could improve the activity of ALP while had nothing to do with the proliferation. With higher concentration of rhbFGF(50 ng/ml).Conclusion: rhbFGF and rhBMP-2 may have synergistic effect on bone formation.The function may be modified by the concentrations of both growth factors. The effect was the strongest for the proliferation of rabbit osteoblast-like cells with 10 ng rhbFGF and 100 ng rhBMP-2/ml.
