摘要
目的:构建用于防治仙台病毒感染的核酸疫苗并初步鉴定。方法:使用逆转录-聚合酶链反应(RT鄄PCR),从新分离的仙台病毒天津株基因组中扩增出HN全部编码序列,将其插入pcDNA3的Pcmv下游,构建仙台病毒HN基因核酸疫苗。PCR、酶切和测序进行鉴定。结果:鉴定证实了核酸疫苗的构建。结论:构建好的核酸疫苗为下一步的体外表达和动物免疫实验奠定基础。
Objective: A specific DNA vaccine has been constructed in order to prevent human being from parainfluenza family infection. Methods: HN-coding sequence has been amplified from new isolated Sendai virus, tianjin strain using RT-PCR, and inserted into the downstream of Pcmv of vector pcDNA3, therefore, DNA vaccine harboring HN gene of Sendai virus is constructed. The recombinator is well identified by series of methods such as PCR, RE digestion and sequencing. Results: The construction has been successfully verified by the methods mentioned above. Conclusion: The recombinator will provide framework for its expression in vitro and animal immunological test in the future.
