摘要
[目的 ]探讨肠复康中药对人结肠癌细胞HT 2 9增殖及浸润转移影响的分子机制。 [方法 ]采用体外培养 ,应用免疫组化方法 ,检测其对核增殖抗原Ki 6 7、金属基质蛋白酶 (MMP2 )及其相应组织抑制物 (TIMP2 )表达的影响。 [结果 ]经中药血清处理过的HT 2 9细胞表现出增殖受抑 ,MMP2 表达减少及TIMP2 分泌增加 ,且其表达与中药血清浓度有量效关系 ,以 30 %血清组效果最明显。 [结论 ]中药肠复康通过抑制Ki 6 7的表达及影响MMP2 、TIMP2 的相互平衡来影响结肠癌细胞的增殖及浸润转移。
Objective] To explore the effect of changfukang recipe on proliferation and infiltration of human colorectal carcinoma cell HT 29 and its mechanism. [Methods] Cell culture and immunohistochemistry were applied to detect the expression of nuclear product antigen ( Ki 67), matrix metalloproteinases 2( MMP 2 ) and its tissue inhibitor (TIMP 2). [Results] The expression of Ki 67, MMP 2, TIMP 2 were observed following the treatment by drug serum of changfukang. The expression of both Ki 67 and MMP 2 decreased while the excretion of TIMP 2 increased, which showed dosage dependent. The effect of 30% changfukang serum on carcinoma cells was the most obvious.. [Conclusion] Proliferation and infiltration of colorectal carcinoma cells could be inhibited by changfukang recipe and its mechanism might be related with inhibiting the expression of nuclear product antigen Ki 67 and affecting the mutual balance between MMP 2 and TIMP 2 .
