摘要
目的 探讨溶酶体中CathepsinB与扩张型心肌病发病的关系。 方法 收集我院因终末期扩张型心肌病行心脏移植术的受体标本 2 0例为实验组及脑死亡 3h内排除心脏疾患的心脏标本 5例为正常对照组 ,通过免疫组织化学 ,RT -PCR和WesternBlotting等方法自不同水平对CathepsinB进行检测。 结果 发现两组心肌组织中的CathepsinB均有阳性信号 ;RT PCR半定量检测CathepsinBmRNA的表达示扩张型心肌病组CathepsinB与 β actin条带面积积分比值明显高于对照组 ,分别为 0 .92 6± 0 .30、0 .4 4 2± 0 .2 5 ,两组间有显著差异 (P <0 .0 1) ;同时从蛋白水平检测CathepsinB的表达 ,显示扩张型心肌病组CathepsinB与 β actin条带面积积分比值明显高于正常组 ,分别为 0 .4 5 4± 0 .0 8及 0 .131± 0 .0 3,两组间有显著差异 (P <0 .0 1)。结论 与对照组相比 ,扩张型心肌病心肌标本中CathepsinBmRNA与蛋白质的表达量均明显增高 ,提示CathepsinB可能参与了扩张型心肌病的发生、发展过程 。
Purpose: To observe the relationship between lysosomal protease cathepsin B and dilated cardiomyopathy( DCM). Methods: Myocardium tissue specimens derived from 20 patients underwent orthotopic cardiac transplantation and 5 normal myocardium samples were collected autopsy from sudden death as controls. In those myocardium cathepsin B was detected by immunohistochemistry, RT-PCR and Western Blotting, respectively. Results: Immunohistochemistry results showed that the positive signals could be found in the myocardium of DCM, and control group also. However, the expression of cathepsin B mRNA in transplantation myocardium markedly statistically increased, compared with that in control group. Meanwhile, the expression of cathepsin B protein also showed significantly higher in DCM group than that in control group by western blotting. Conclusions: It is presumed that cathepsin B participates in the genesis and development of DCM, and that cathepsin B may play an important role in the myocardial apoptosis in DCM.
