摘要 目的 探讨血小板源性生长因子 (PDGF)及受体的表达与子宫肌瘤的形成和发展的关系。方法 对 2 1对子宫肌瘤和邻近正常平滑肌的石蜡组织切片进行免疫组织化学染色 ,检测PDGF AA、PDGF BB、PDGFR α和PDGFR β的表达情况。结果 PDGF AA、PDGF BB及PDGF受体α、β在肌瘤组织中的表达量均较邻近正常平滑肌组织显著增高 (P <0 .0 5 ) ;其中PDGF BB的表达受月经周期的影响 ;子宫肌瘤组织中 ,PDGF BB的表达比PDGF AA高 (P <0 .0 1) ;PDGFR α的表达较PDGFR β高 (P <0 .0 1)。结论 PDGF及其受体在子宫肌瘤组织中局部的高表达可能与子宫肌瘤平滑肌细胞的高增殖状态有关 ,PDGF BB可能作为主要因子参与卵巢激素刺激的子宫肌瘤平滑肌细胞的增殖 ,而PDGFR α则作为主要的受体类型介导PDGF在子宫肌瘤发生。 Objective To study the expression of platelet-derived growth factor (PDGF) and the receptors in human uterine leiomyoma and their possible roles in the formation and progression of the leiomyoma.Methods Myometrium and leiomyoma tissue pairs were obtained from 21 women who underwent hysterectomy in different phases of menstrual cycle. The paraffin embedded samples were stained by immunohistochemical methods. The expression of PDGF-AA, PDGF-BB, PDGFR-α and PDGFR-β were detected.Results The expression of PDGFs and the receptors were significantly higher in leiomyoma than in myometrium (P< 0.05). There was significant difference in the expression of PDGF-BB between proliferative and secretory phases of menstrual cycle. Interestingly, it was also found that in leiomyoma tissue, the level of PDGF-BB was higher than that of PDGF-AA (P< 0.01), and the level of PDGFR-α was higher than that of PDGFR-β (P< 0.01).Conclusion The higher expression of PDGFs and their receptors in local tissues of leiomyoma indicated that the autocrine and paracrine mechanism of the PDGFs might be involved in the formation and the progression of uterine leiomyoma.
