摘要
目的:探讨改进后的线栓法大鼠永久性大脑中动脉梗塞(PMCAO)局灶性脑缺血模型的使用价值。方法:分离颈总动脉、颈内动脉、颈外动脉,结扎颈外动脉,经由颈总动脉剪一小口,将制备好的栓线插入颈总动脉,并推进送入颈内动脉内,阻断大脑中动脉,制备成PMCAO模型,并观察大鼠脑组织的病理改变。结果:成功率可高达95%,模型符合脑梗死病理过程。结论:改进后的方法(PMCAO)简便、易重复,降低了动物死亡率,提高了成功率,可做为缺血性脑血管病研究的理想动物模型。
Objective To study the worth of permanent middle cerebral artery occlusion(PMCAO)improved from the classic su-ture model(Longa et al,1989).Method Briefly,the right common carotid artery(CCA)and the bifurcation of the right internal carotid artery(ICA)and external carotid artery(ECA)were surgically exposed.A6suture was tied loosely around the ECA,and a microvascular clip was placed on the CCA and the ICA adjacent to the ECA origin.A5cm length of 4monofilament nylon suture,its tip rounded by heating near a flame,was introduced into the ECA through an incision.A6suture was secured around the ECA stump,to prevent bleeding through the incision.The microvascular clip was removed and the suture was gently advanced from the E-CA to the ICA.The path of the suture toward the base of the skull was visualized.Approximately17.5-18mm of suture was inserted.Insertion continued until resistance was felt and curving of the suture and mild stretching of the ICA was seen,indicating that the su-ture tip has passed the MCA origin and reached the anterior cerebral artery(ACA),which has a smaller diameter.Result To use this method,95%of them are successful.Conclusion it's a perfect method to make PMCAO.
