摘要
目的探讨内源性糖皮质激素(EGC)与酒精性股骨头缺血坏死骨细胞凋亡以及相关蛋白p53和Bcl-2的关系.方法家兔90只,随机分为三组,灌胃法建模,实验组予含50%乙醇的烈性白酒8ml·kg·d-1,干预组给予氨鲁米特25mg·kg·d-1,对照组只给等量生理盐水.1~6个月分批处死,检测血清中EGC浓度、骨细胞凋亡、p53和Bcl-2蛋白的表达.结果实验组血清EGC浓度高于干预组和对照组(P<0.01).2个月时起实验组骨细胞凋亡数明显高于干预组和对照组,但干预组与对照组之间差异无显著性(P>0.05);3、6个月时,凋亡细胞逐渐增多,三组间差异均有显著性(P<0.05).第2个月时骨细胞Bcl-2和p53蛋白阳性细胞数,实验组与对照组之间差异有显著性(P<0.05),干预组与实验室和对照组之间差异无显著性;3、6个月时,三组间差异均有显著性(P<0.05).结论过量饮酒后所致的EGC浓度增加使p53蛋白上调和Bcl-2蛋白下调是引起股骨头骨细胞凋亡的重要原因.
Objective To investigate the effect of the endogenous glucocorticoid(EGC) and the expression of p53 and Bcl-2 proteins of osteocyte apoptosis in avascular necrosis of the femoral head was induced by alcoholism. Methods 90 rabbits were divided into three groups randomly. The experimental group were administered with the 50% contained spirit of alcohol at dosage of 8 ml·kg-1·d-1, the intervention group were given aminoglutelhimide 25 ml·kg-1·d-1, and the control group were given isodose NS only by pouring into stomach with tube. The levels of serum EGC were detected from 1 to 6 months. The osteocytes apoptosis was measured by means of TUNEL, and the expression of p53 and Bcl-2 proteins by immunohistochemistry. Results The levels of serum EGC in the experimental group were higher than that in the intervention group and control group(P<0. 01). The numbers of apoptosis osteocytes in the experimental group were higher than that of the intervention group and the control group (P<0. 01),in third and sixth month. Insecond month, the amount of Bcl-2 protein expression of osteocytes in the experimental group decreased, and the level of p53 protein of osteocytes were increased than that in other groups. In third and sixth month, the amount of Bcl-2 protein expression of osteocytes in the experimental group significantly were decreased, the expression of p53 protein of osteocytes were remarkably increased than that in other groups (P<0. 05). Conclusion EGC was an important factor in the apoptosis in osteocytes of ANFH by alcoholism.
