摘要 目的:探索骨髓基质细胞黏附特征的物质结构基础,为分离鉴定骨髓基质细胞提供依据。方法:实验于2004-07在珠江医院神经外科实验室进行,对原代培养的兔骨髓基质细胞,在第1,7,14天进行原子力显微镜观察。结果:骨髓基质细胞在不同的培养阶段都具备黏附特性,从而表现为贴壁生长,细胞周缘均可见到条纹状结构,且大多呈明暗相间规则有序的排列,少部分存在交叉。该条纹状结构较平直,表面光滑,远端插入细胞周围基质的下方。结论:原子力显微镜适于对细胞表面的超微结构观察;骨髓基质细胞的黏附特征具有相应的形态结构。 AIM:To investigate the basis of material structure of adhesion characteristics of bone marrow stromal cells(BMSCs),and provide evidence for the isolation and differentiation of BMSCs. METHODS:The research was carried out in the Laboratory of Neurosurgery Zhujiang Hospital from July to August 2004.The rabbit BMSCs in primary culture were scanned with atomic force microscope (AFM)on days 1,7 and 14. RESULTS:Adhesion was the characteristics of BMSCs in the different stages of culture.And all the cells adhering to the wall were surrounded by the structure of stripes. These stripes were the alteration of bright and dark and arranged in order.Only a few were crossed.The stripes were straight and smooth. And their terminals were inserted into the extracellular matrix. CONCLUSION:AFM has special predominance in observation of the ultrastructure of cell surface. BMSCs have adhesion characteristics with corresponding structure.
