中子及γ线照射小鼠肠道损伤的定量病理研究 被引量:1

QUANTITATIVE PATHOLOGIC STUDIES ON THE INJURIES OF MICE INTESTINES INDUCED BY NEUTRON AND γ RAYS IRRADIATION

摘要 目的 定量比较研究小鼠经中子及γ线照射后肠道损伤的病理特点。方法 350只二级雄性BALB/C小鼠,经不同剂量的中子和γ线照射,于照后6h、12h、1d、2d、3d、4d、5d、7d、10d、14d、21d及28d分批活杀,进行全肠长度及重量测量;全肠组织10%缓冲福尔马林固定,石蜡包埋制片, 潘氏细胞及HE染色,Leica图像分析仪检测隐窝细胞数密度和隐窝内潘氏细胞的面密度。结果 中子及γ线照射后全肠的长度缩短、重量减轻,且重量减轻先于长度缩短。2.5Gy中子照射后肠粘膜见明显损伤及损伤后恢复现象,4.0Gy以上照射未见明显恢复。g 射线5.5Gy照射未见粘膜剥脱,12Gy组隐窝再生能力介于中子4.0-5.5Gy之间。中子2.5Gy照后1d内,隐窝细胞数进行性减少,于照后3d增多,7d达高峰,而5.5Gy中子和12Gyγ线于照后3d内均明显进行性减少(p<0.01)。γ线5.5Gy隐窝细胞增加发生时间早,高峰提前。中子2.5Gy照后7d时潘氏细胞明显增加(p<0.05);而5.5Gy组于照后2d潘氏细胞相对增多(p<0.05)。5.5Gyγ线于照后5d增加(p<0.05)。结论 照射后肠隐窝细胞明显损伤,相同剂量照射后,中子对隐窝细胞损伤明显重于γ线;射线对潘氏细胞影响较小,相同剂量的中子及γ线照射后,潘氏细胞出现不同的变化规律。 Objective To study quantitatively and comparatively the pathologic characteristics of mice intestinal injuries induced by neutron andγrays irradiation. Methods 350 male BALB/C mice were used and irradiated with neutron andγ rays, sacrificed at 6 hours,12hours,1day,2 days,3 days,4 days,5 days,7 days,10 days,14 days,21 days and 28 days respectively after irradiation. The length and weight of whole gut were detected and the guts were fixed in 10% buffer formallin and cut paraffin section, Paneth抯 cell and H&E staining, and observed by LM. The number density of crypt cell and the area density of Paneth抯 cell were detected by means of LEICA image analysis. Results The length became short and the weight decreased for the irradiated gut. Furthermore, decreased weight was earlier than shortened length. The intestinal mucosa could be seen injury and then recovery obviously in 2.5Gy neutron group, while over 4.0Gy radiation not recovery. The mucosa was not been denudation by 5.5Gy g -rays irradiation. The regeneration capabilities of crypt cell were between 4.0 and 5.5Gy in 12Gy g-rays. The numbers of crypt cells were decreased progressively within 1 day after 2.5Gy neutron irradiation, increased on day 3 and at peak on day 7, while the crypt cell was progressively decreased within 3 days after 5.5Gy neutron and 12Gyγrays irradiation. The time and the peak of crypt cells regeneration in 5.5 Gyγ-rays were earlier than that of 2.5Gy neutron. The Paneth抯 cell was increased on day 7 in 2.5Gy neutron group, relative increase day 2 in 5.5Gy neutron group, and also increased on day 5 in 5.5Gy γ-rays group. Conclusion The gut crypt cell occurred obvious injury postirradiation, furthermore, the injury of crypt cell was severe obviously in neutron group at the same dose; The Paneth抯 cell was less influence postirradiation, and had different changes after neutron and g-rays irradiation.
