摘要
目的 用CD34抗原标记法定性及定量研究兔实验性牙齿移动过程中牙周组织的血管改建规律。方法 35只大耳白兔随机分为 7组 ,即正常组及加力 1、3、5、7、14、2 1天组 ,每组 5只。麻醉下于上颌第一磨牙至相应切牙间拴结不锈钢螺簧 ,施力 80 g左右。对组织学标本进行CD34(血管内皮细胞标记物 )免疫组化染色 ,光镜下观察兔牙周组织血管改建的变化。应用图像分析系统测定牙周组织微血管密度 (MVD)与微血管面积 (MVA)。结果 图像分析结果显示 ,压力区与张力区MVD的时间变化规律基本一致 ,峰值均出现于加力 7天组 ,分别为 13.0 3± 8.6 0个 /mm2 与 13.5 8± 8.2 9个 /mm2 ;压力区与张力区MVA的时间变化规律亦基本相似 ,峰值均出现于加力第7天 ,分别为 2 4 2 6 .6 3± 6 4 0 .2 0 μm2 与 2 2 17.5 3± 4 4 3.0 4 μm2 。结论 在兔实验性牙齿移动过程中 ,压力区与张力区牙周组织血管改建的时间变化规律基本一致 ,血管新生的峰值均出现于加力第 7天。
Objective To study the regularity of blood vessel remodeling(BVR) of rabbit periodontal tissue during experimental tooth movement qualitatively and quantitatively by using antibody against CD34.Methods Thirty-five rabbits were divided into normal group and experimental (1,3,5,7,14,21day) group of 5 rabbits each.After anesthesia,a coil spring was deposited between the maxillary first molar and the corresponding central incisor,the force was almost 80g.All of the tissue sections were proceeded with antibody against CD34 (vascular endothelial cell marker) immunohistochemical staining,and the changes of BVR of rabbit peridontal tissue was observed under the light microscope(LM).Microvessel density(MVD) and microvessel area(MVA) of periodontal tissue were measured by Computer Image Analyzing System,the regularity of the changes of BVR was studied.Results The results of image analyzing indicated that the regularity of change of MVD at the pressure zones was agreement with that at the tension zones,all peak occurred in the 7th days group,and were 13.03 ± 8.60/mm 2 and 13.58 ± 8.29/mm 2 respectively;the regularity of change of MVA at the both zones was almost similar,all peak also occurred in the 7th days group,and were 2426.63 ± 640.20μm 2 and 2217.53 ± 443.04μm 2 respectively.Conclusion The regularity of change of BVR of periodontal tissue at the pressure zones and the tension zones during experimental tooth movement was almost similar,the angiogenesis peak all occurred in the 7th days group.
