白细胞介素lra基因多态性与2型糖尿病关系的研究

摘要 目的 探讨白细胞介素1受体拮抗剂(IL-lra)基因多态性与2型糖尿病的关系。方法 应用聚合酶链反应技术对109例2型糖尿病患者和201例正常人的白细胞介素1受体拮抗剂基因(IL一1RN)数目可变的串联重复多态性进行分析。结果 2型糖尿病患者和正常人IL-1RN Ⅰ/Ⅰ、Ⅰ/Ⅱ、Ⅱ/Ⅱ和Ⅰ/Ⅳ4种基因型频率分别为80.7%和81.1%,14.7%和16.9%,1.8%和0.5%,2.8%和1.5 9%;IL-1RN*Ⅰ等位基因频率分别为89.5%和90.3%,IL-1RN*Ⅱ等位基因频率分别为9.1%和9.0%。携带IL-1RN*Ⅱ等位基因的2型糖尿病组患者血糖和甘油三酯水平明显升高。结论 IL-1RN内含子2基因多态性与2型糖尿病易感性无关,但IL-1RN(Ⅱ)等位基因可能与2型糖尿病病情的危险性有关。 Objective To determine the allele frequencies and genotype distribution of IL-1 receptor antagonist (IL-lra) gene (IL-1RN) polymorphism in type 2 diabetes mellitus patients. Methods Using polymerase chain reaction (PCR) to analyze the polymorphism of variable number of tandem repeat (VNTR) of IL-lra gene among 109 patients with type 2 diabetes mellitus and 201 controls in the present study. Results The frequencies of Ⅰ/Ⅰ,Ⅰ/Ⅱ,Ⅱ/Ⅱ and Ⅰ / Ⅳ were 80. 7%, 14. 7%,1.8% and 2. 8% in type 2 diabetes mellitus patients respectively,while in controls were 81. 1%,16. 9%,0. 5% and 1. 5%. The IL-1RN * Ⅰ allele was detected in 89. 5% of type 2 diabetes mellitus patients and 90. 3% of controls! while the IL-1RN * Ⅱ allele was detected in 9. 1% of patients and 9. 0% of normal controls. The levels of blood glucose and triglycerides of with IL-1RN * Ⅰ allele were higher than that of type 2 diabetes mellitus without IL-1RN * Ⅱ allele. Conclusion The polymorphisms of VNTR of IL-lra are not associated with the susceptibility of type 2 diabetes mellitus. Carrying IL-1RN * Ⅱ allele could increase the risk of type 2 diabetes mellitus.
