摘要
栀子经脱脂热水提取,Sevage 法脱蛋白质,乙醇沉淀得粗制品多糖 GPS1.经 DEAE—纤维素(DE—52)层析得 GPS2和 GPS3 2个组分.分别经 Sephadex G—200层析进一步纯化得精品GPS_4和 GPS_5.玻璃纤维纸电泳、乙酸纤维薄膜电泳和琼脂糖电泳均显示单一条带.GPS_4和 GPS_5不含蛋白质和核酸物质.用 SepbadeX G—150层析法测得 GPS_4和 GPS_5的平均分子量分别为14000和10000.体外试验表明此多糖对肉瘤 S—180和腹水肝癌细胞生长均有一定的抑制作用.
Two polysaccharides,named GPS_4 and GPS5,are isolated fromgardenia jasminoides ellis.The crude polysaccharide is obtained by extractionwith boiling water,by deproteinization with sevage method,by precipita-tion with ethanol.The crude product is taken up in a DEAE-cellulose(DE-52)column.GPS_4 fraction is isolated by eluting with water,and GPS_5 by a linear gradient(o—4mol/L).Both fractious are furtherpurified by chromatography with gel filtration(Sephadex G-200).Bothfractions show chemical homogeneity by means of agarose electrophoresis,cellulose acetate membrane electrophoresis,glass-filter paper electrophoresis.Neither GPS4 nor GPS_5 contains protein and nucleic acid.The averagemolecular weights of GPS4 and GPS_5 are estimated to be approximately1.4×10~4 and 1×10~4,respectively.The experimentation in vitro indicatesthat this polysaccharide shows an evident inhibitory activity on thecells of the implanted tumor sarcoma 180 and ascites hepatoma.
