通过流式细胞术(FACS),利用细胞表面糖蛋白(Jlld.2)单克隆抗体研究了J l ld在新制备的小鼠脾脏B细胞上的表达,特别是细菌脂多糖(lipopolysaccharide,LPS)刺激后Jl ld表达的变化。发现浮力密度较高的小B细胞高度表达Jl ld(Jlld~高),而Jl ld低细胞则为浮力密度较低的大B细胞。用LPS刺激脾脏高浮力密度小B细胞,可诱导Jl ld表达的降低。这些结果表明,B细胞活化后Jl ld的表达降低,Jl ld的高和低并不是一个稳定的细胞族系(lineage)标志。
The expression of Jlld on mouse splenic B cells and its change following B cell activation were studied by using FACS. The results showed that the high-density small splenic B cells had a phenotype of Jlld^(high) and Jlld^(low) B cells fell into low-density large B cells. Activation of B cells by different stimulators produced differences in the level of expression of Jlld. Stimulation of B cells by LPS led to decrease in Jlld, but Jlld remained high following anti-IgM stimulation. Additional stimulation of IL-6 following anti-IgM stimulation decreased the expression of Jlld. Our experiments do not directly address whether low Jlld is a marker of memory B cell lineages, but they do indicate that the expression of Jlld decreases following activation and high'and low surface Jlld are not stable lineage markers.
