摘要 目的 观察治疗量红霉素与钙拮抗药合用对动物胃肠运动的影响。方法 采用整体实验研究治疗量红霉素与硝苯地平合用对小鼠胃排空和肠内容物推进的影响 ;采用离体方法研究红霉素对兔回肠平滑肌收缩活动的影响以及地尔硫卓对红霉素作用的影响。结果 治疗量红霉素明显减慢小鼠的胃排空和肠内容物推进 ,与较大剂量硝苯地平合用时小鼠的胃排空和肠内容物推进进一步减慢 ;离体试验结果表明 ,5× 10 - 6 ~ 5× 10 - 5mol·L- 1 红霉素可引起兔回肠平滑肌与浓度相关的收缩活动增强 ,该作用可被地尔硫拮抗。结论 治疗量红霉素明显减慢小鼠的胃排空和肠内容物推进 ;与较大剂量硝苯地平合用时该作用进一步增强 ;红霉素明显增强兔离体回肠平滑肌的收缩活动 ,该作用与促进细胞外Ca2 Objective: To observe the effects of the therapeutic dose of erythromycin in combination with calcium antagonists on gastrointestinal motion in mice and rabbits Methods: The effects of erythromycin in combination with nifedipine on gastrointestinal motion in mice were studied in vitro The effects of erythromycin alone and in combination with diltiazem on contractile activity of isolated ileal smooth muscle in rabbits were investigated in vitro Results: Erythromycin decreased significantly the velocity of gastric emptying and prochoresis of intestinal contents in mice, and more so in mice which received erythromycin in combination with a large dose of nifedipine In vitro experiment, 5×10 -6 ~5×10 -5 mol·L -1 erythromycin evoked the contractions of ileal smooth muscle in rabbits in a dose dependent manner, and this effect was inhibited absolutely by 10 -5 mol·L -1 diltiazem Conclusion: A therapeutic dose of erythromycin in combination with nifedipine inhibits gastric emptying and prochoresis of intestinal contents in mice in a greater degree than that of erythromycin alone In addition, erythromycin enhances the contractile activity of isolated ileal smooth muscle in rabbits, which may be exerted by increasing the inflow of extracellular calcium
