摘要
本文用氯仿处理,PEG沉淀;超离,Sepharose 4B柱层析及蔗糖密度梯度离心等方法纯化了兔瘟病毒(RPeV)。电镜下观察到RPeV为无囊膜,大小为32~34nm。纯化了的RPeV通过SDS-聚丙烯酰胺连续梯度凝胶电泳进行多肽分析。发现RPeV多肽只有1条63~64kd主要结构蛋白组成。用SPF兔肝粉吸收后的抗RPeV抗体作免疫转印。证明这条多肽是RPeV所特有的。
Rabbit pest virus (RPeV) was isolated from infected rabbit livers by means of treatmentwith chloroform, PEG precipitation and differential centrifugation. The sepharose 4B cloumnchromatography and linear sucrose gradient centrifugation was also used in further purification. Thevirons were icosahedral, non-enveloped, 32~34nm in diameter in negative staining preparation. TheRPeV virons contained only one major structural protein with a molecular wight of approximately 63~64kd by SDS-PAGE and immunoblot with scra against RPeV absorbed with SPF rabbit liver powder.
