摘要
目的 用离体蟾蜍肾上腺髓质嗜铬细胞建立刺激 分泌偶联的模型。方法 用胶原酶 (0 5mg ml)消化蟾蜍肾上腺 ,再用牛血清白蛋白 (5mg ml)终止消化 ,获得离体的蟾蜍肾上腺髓质嗜铬细胞。用乙酰胆碱 (2 0 0μmol L)或高K+ (6 5mmol L)刺激离体嗜铬细胞分泌儿茶酚胺。结果 消化培养后获得了离体的蟾蜍肾上腺髓质嗜铬细胞。ACh(2 0 0 μmol L)能刺激离体细胞分泌CA ,回归方程为Y =- 16 12 +16 94X ,r=0 995 1>α0 0 1 =0 95 87;K+(6 5mmol L)也能刺激离体细胞分泌CA ,回归方程为Y =12 5 3+7 933X ,r =0 994 2 >α0 0 1 =0 95 87。结论 离体的蟾蜍肾上腺髓质嗜铬细胞是理想的刺激 分泌偶联模型。由ACh和高K+ 促进的嗜铬细胞CA分泌具Ca2 + 浓度的依赖性。
Objective To establish the ideal model of stimulus-secretion coupling by isolated adrenal chromaffin cells of the toad. Method The toad adrenal gland was digested by collagenase (0.5mg/ml) and cultivated a short time in Ringer containing bovine serum ablumin (5 mg/ml). Then chromaffin cells isolated were stimulated by adding ACh (200 μmol/L) or excess K++ (65 mmol/L) to induce the CA secretion. Results Isolated toad adrenal medullary chromaffin cells were obtained. The ACh (200 μmol/L) induced the toad chromaffin cells to release CA. The regression equation was Y=-16.12+16.94X,r=0.9951>α 0.01 = 0.958 7 . The K++(65 mmol/L) also induced the cells to release CA, and the regression equation was Y=12.53+7.933X, r=0.9942>α 0.01 =0.9587. Conclusion Isolated toad chromaffin cell can be used as an ideal model for studing stimulus-secretion coupling. The stimulation of ACh and excess K++ is dependent on the extracellular Ca+{2+} concentration.
