反义寡核苷酸技术及其在NMDA受体亚单位研究中的应用

摘要 反义寡核苷酸技术是利用碱基互补配对的原则 ,设计一段寡脱氧核苷酸 ,与靶序列特异结合 ,从而抑制基因表达 ,降低mRNA和 /或蛋白质水平 ,影响动物的后续生物效应。反义寡核苷酸技术作为一种快速、特异性阻断基因表达的方法 ,近二十年来取得迅速发展 ,其潜在应用价值巨大。特别是近年来在对NM DA受体领域的反义研究 ,越来越得到人们的重视。本文就反义寡核苷酸技术及当前反义寡核苷酸技术在NMDA受体亚单位研究中的应用进行阐述。 On the basis of the base complementation and pair-matching, a little length section comprised twelve to thirty bases is designed. Those bases aim at the target sequence coding specific functional protein which has definite ability to mediate physiological function or pathologic injury progress. By binding to the target sequence, they suppress the gene duplication, transcription and translation, showing off the lower level of the mRNA and/or the protein, furthermore influencing on the successive biological effects. The antisense technique has developed fast in the past twenty years for the quick and specific gene block and its latent application value seems considerable. Especially the research on the N methyl D aspartate receptor (NMDAR) subunits by the antisense oligodeoxynucleotide technique has received much recognition. The article is to review the antisense technique and its use in the research on the NMDAR subunits.
