摘要
为了探讨人类运动能力的相关基因标记与分子遗传学机制 ,选取汉族耐力运动员 61人 ,相应汉人对照 63人 ,对其线粒体基因高变区I(mtDNAHVRⅠ )特异性片段进行扩增、测序 ,评估其单倍型、多样性及偶合概率 ,并与其他人群进行比较研究。结果显示 :中国汉族耐力运动员中有60个单倍型 ,汉人对照有 61个单倍型。中国汉族耐力运动员mtDNA多样性高达 99 95 % ,汉人对照mtDNA基因多样性也达 99 90 % ,二组无关个体间偶合概率均很小 ,经卡方检验上述各参数与频率在汉族耐力运动员和汉人对照两组间的分布无显著性差异 (P >0 0 5 )。与西班牙人、中国台湾人及俄罗斯人等不同人群比较发现 ,mtDNA多样性的差异存在于群体内个体间 ,而群体间的差异较小。结果提示 ,汉族耐力运动员mtDNAHVRⅠ多样性非常高 ,且每个个体间mtDNA单倍型差异很大 ;不同人群mtDNA多样性的差异存在于群体内个体间 ,而群体间的差异较小。
In order to investigate association between gene marker and human athlete performance and their molecular genetic mechanism, we analyzed the sequence polymorphisms of the mitochondrial DNA hypervariable regionⅠ(mtDNA HVRⅠ). All subjects were Han nationality, including 61 endurance athletes and 63 healthy controls. Sequence polymorphisms of HVRⅠwas determined by polymerase chain reaction (PCR) and direct sequencing. The results showed that there were 60 haplotypes in Chinese endurance athletes and 61 haplotypes in healthy controls. The genetic diversity of Chinese endurance athletes was 99.95%, and 99.9% for the controls.The coincident probability between two individuals was very little in both groups. As compared with other populations, the differences of genetic diversity appeared in individuals rather than in populations. The results suggested that Chinese endurance athletes and their controls showed a high mtDNA diversity and the differences of genetic diversity appeared in different individuals of one population rather than different populations.
