摘要
为探讨极低密度脂蛋白受体在摄取富含甘油三酯的脂蛋白及泡沫细胞形成中的作用 ,在体外实验中 ,将极低密度脂蛋白、β极低密度脂蛋白及低密度脂蛋白分别与小鼠腹腔巨噬细胞孵育 2 4及 4 8h后 ,检测细胞内甘油三酯及总胆固醇的含量 ,油红O染色观察泡沫细胞的形成 ,半定量逆转录聚合酶链反应检测极低密度脂蛋白受体、LRP及极低密度脂蛋白受体的mRNA表达变化。结果发现 ,三种脂蛋白与巨噬细胞均能升高细胞内甘油三酯、总胆固醇含量 ;极低密度脂蛋白受体受极低密度脂蛋白、β极低密度脂蛋白的刺激表达上调 ,低密度脂蛋白受体表达下调 ,LRP表达略有上调。在稳定表达极低密度脂蛋白受体细胞中的实验表明 。
Aim To discuss the function of very low density lipoprotein (VLDL) in foam cell formation and the role of VLDL receptor (VLDLR) in this process. Methods Macrophage cells separated from mouse abdominal cavity were incubated with three lipoproteins, VLDL, β-VLDL, and low density lipoprotein (LDL) for 24 h, 48 h respectively. The content of triglyceride (TG) and total cholesterol (TC) in cells was examined, and the foam cells formation was identified by oil red O dying. The mRNA expression level of LDLR, LRP and VLDLR was measured by half-quantitation RT-PCR. Results The content of TG and TC increased by all three lipoprotein; VLDLR was upregulated by VLDL and β-VLDL while LDLR was downregulated and LRP was upregulated a little. Conclusion The experiment in the ldl-a7-VR cells which can steadily express VLDLR shows that the uptake of triglyceride-rich lipoprotein (TRL) mediated by VLDLR plays an important role in the accumulation of lipids and the foam cell formation.
