摘要 目的观察Pitavastatin对血管内皮细胞VEGF、bFGF等因子表达的影响,探讨他汀类药物可能具有的独立于血脂调节作用以外的促血管新生作用。方法体外培养HUVEC,加入不同浓度的Pitavastatin后用蛋白印迹杂交法检测VEGF,bFGF,p-Akt及p-eNOS蛋白表达水平;镉还原Griess测定培养基中NO含量。结果在培养的HUVEC中加入Pitavastatin后,VEGF,bFGF,p-Akt及p-eNOS蛋白呈现高表达,培养基中NO含量明显增高。结论Pitavastatin能诱导内皮细胞VEGF,bFGF,p-Akt及p-eNOS蛋白的表达及增加NO含量,推测其可能通过VEGF-p-Akt-eNOS(NO)径路而发挥非调脂的作用。 Objective: To observe the influence of pitavastatin on expression of intercellular vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) in human umbilical vein endothelial cells (HUVEC )and investigate the non-lipid mechanisms of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitors on angiogenesis. Methods: The expression of VEGF, bFGF, p-Akt and p-eNOS in protein level was detected by western blot, and level of serum NO production was done by Griess in HUVEC co-culture. Results: Pitavastatin treatment up-regulated protein expression of VEGF, bFGF, p-Akt and p-eNOS, and serum NO production was enhanced markedly in the pitavastatin. Conclusions: The expression of VEGF, bGFG, p-Akt and p-eNOS in HUVEC suggests a novel function of pitavastatin as a positive modulator of vessel growth with VEGF-p-Akt-eNOS (NO) pathway.
