摘要
目的 观察烟雾吸入伤大鼠肺组织抗氧化酶活性和谷胱甘肽含量变化。方法 建立大鼠烟雾吸入伤模型 ,将 6 0只Wistar大鼠随机分成正常对照组 ,烟雾吸入伤 1、3、6、12h和 2 4h组 ,分取肺组织测定过氧化氢酶 (CAT)、谷胱甘肽过氧化物酶(GSH PX)、超氧化物歧化酶 (SOD)活性和谷胱甘肽 (GSH)含量。结果 肺组织匀浆CAT活性在烟雾吸入伤后 3、6h明显高于正常对照组 (P <0 .0 5、P <0 .0 1) ,伤后 12h、2 4h已低于正常对照组 ;GSH PX活性伤后 1h已明显高于正常对照组 (P <0 .0 1) ,伤后3h达峰值 ,伤后 12、2 4h明显低于正常对照组 (P <0 .0 5 ) ;SOD活性和GSH含量从烟雾吸入伤后 1h开始明显低于正常对照组 ,一直持续到伤后 2 4h(P <0 .0 5 ,P <0 .0 1)。结论 烟雾吸入伤后肺组织CAT、GSH PX活性先增后减 ,而SOD活性和GSH含量持续降低 ,反映了肺内氧应激的强度和抗氧化物质的不同作用。
Objective To observe the changes in the activities of antioxidases and content of glutathione of lung tissue in rat after smoke inhalation injury.Methods The model of smoke inhalation injury in rat was set up,sixty Wistar rats were randomly divided into six groups such as control group ,and the groups of 1h,3h,6h,12h and 24h following smoke inhalation.The lung tissue was isolated, the activities of catalase(CAT), glutathione peroxidase(GSH-PX) and superoxide dismutase(SOD) and the content of glutathione in lung tissue homogenate were assayed.Results CAT activity in lung tissue homogenate in groups of 1h and 3h following smoke inhalation were significantly higher than it in control group.GSH-PX activity in lung tissue homogenate in groups of 1h and 3h following smoke inhalation were markedly higher than it in control group,but the activity in groups of 12h and 24h following smoke inhalation were obviously lower than it in control group.SOD activity and the content of glutathione in lung tissue homogenate were remarkably decresed from 1h to 24h while comparing with control group.Conclusion The activities of CAT and GSH-PX in lung tissue homogenate after smoke inhalation injury are increased in the early stage and decreased in the late stage, and SOD activity and glutathione content keep on decline,it reflects the intensity of oxygen stress in lung tissue and the different effects of antioxidants.
