针刺足三里穴对大鼠冷应激性溃疡下丘脑NOS表达的影响 被引量:4

摘要 目的:本实验采用冷应激的方法,研究针刺足三里穴对急性胃溃疡的保护作用,以及应激大鼠下丘脑与应激有关的NOS的表达情况. 方法:应用胃黏膜溃疡指数的计算和RT-PCR的方法研究针刺足三里对大鼠的冷应激性溃疡的保护和下丘脑一氧化氮合成酶(NOS)的表达隋况,经图像分析系统照相并进行半定量. 结果:针刺足三里穴可以明显减少冷直激造成的溃疡指数(正常对照组为0,单纯应激组为26.25±4.40,针刺预防溃疡组为9.75±1.91,针刺预防溃疡组与单纯应激组相比P<0.01).下丘脑NOS1不参与冷应激溃疡病理过程,针刺足三里穴可以在生理范围内上调下丘脑的NOS1的表达过程(正常对照组为0.69±0.05,单纯应激组为0.77±0.09, 针刺预防应激组为1.87±0.15,针刺预防应激组与正常对照组相比P<0.01);下丘脑NOS2参与了冷应激溃疡病理过程,针刺足三里穴可以下调其表达(正常对照组为0.06±0.02,单纯应激组为0.24±0.05,针刺预防应激组为0.12±0.06,P<0.01),减少病理损害程度;NOS3也参与了冷应激溃疡这一病理过程,针刺可以下调其表达作用(正常对照组为0.30±0.08,单纯应激组为0.63±0.14, 针刺预防应激组为0.45±0.14,针刺预防应激组与单纯应激组与正常对照组相比P<0.05,单纯应激组与正常对照组相比P<0.001). 结论:针刺对冷应激溃疡具有保护作用,该种保护作用是通过对生理性上调NOS1的生理性表达,下调NOS2、NOS3的病理性表达而实现的. AIM: To study the protective effect in cold-restraint stress-induced ulcer with acupuncturing the acupoint ST 36, and the expression of NOS related with stress in hypothalamus. METHODS: Ulcer index in rats and RT-PCR were used to study the protective effect in stress-induced ulcer with acupuncture, and the expression of NOS in hypothalamus. Images were analyzed with semi-quantitative methods. RESULTS: The ulcer index was significantly decreased in stress-induced ulcer rats by acupuncturing in contrast with only stress-induced ulcer rats (control group 0.stress ulcer group 26.25 ± 4.40,and acupuncture group 9.75 ± 1.91; Acupuncture group vs stress ulcer group, P<0.01). The expression of NOS1 was increased by acupuncturing protective stress-induced ulcer (0.69 ± 0.05, 0.77 ± 0.09, 1.87 ± 0.15, P <0.01). The increased expression of NOS2 was related with stress-induced ulcer, and acupuncture would decreased its expression (0.06 ± 0.02, 0.24 ± 0.05, 0.12 ± 0.06, P<0.01). The expression of NOS3 had the similar reaction to NOS2, but the decreased effect of acupuncturing was limited (0.30 ± 0.08, 0.63 ± 0.14, 0.45 ± 0.14, P<0.05). CONCLUSION: The protective effects in stress-induced ulcer with acupuncturing the acupoint ST 36 are realized by increase of the physiological expression of NOS1 as well as decrease of the expression of NOS1 and NOS3.
