摘要
研究了猪胰脂肪酶催化拆分丁酸缩水甘油酯的对映选择性水解反应,以拆分得到的酯对映体过量值(ee%)为指标,对有机溶剂与磷酸缓冲液体系中酶催化水解进行了比较研究,并考察了磷酸缓冲液体系中各因素对酶促水解反应中对映选择性拆分效果的影响。结果表明,磷酸缓冲液体系较有机介质转化率高,在磷酸缓冲液体系中缓冲液pH=7.5,底物浓度为0.4mol/L,酶用量为30mg/g(底物),反应时间为4.0h,反应温度为30℃,振荡频率为150r/min的条件下酶促水解的拆分效果最好,对映体过量值达84.65%,此时的转化率为51.78%。
Optical resolution of racemic glycidyl butyrate has been efficient achieved by Porcine Pancreatic Lipase(PPL)catalyzed enantioselective hydrolysis. With the enantiomeric excess ratio (\%ee\%%), the action conditions have been optimized in bufferis. The optimum initial pH of buffer is 7.5. The optimum lipase concentration is about 30 mg/g (butyrate). The optimum initial substrate concentration is around 0.4 mol/L.The optimum temperature for theenzyme reaction is around 30 ℃. The optimum shaking speed is about 150 r/min. The optimum reaction time is about 4 h. Under the condition, we achieved the optical purity of the finally recovered (R)-glycidyl butyrate and the \%ee\%% is about 85%, the substrate conversion efficency is about 51.78%.
