银纳米粒子与血清白蛋白相互作用的滞后效应

摘要 用圆二色光谱,紫外-可见光谱研究银纳米粒子与血清白蛋白的相互作用及其相关效应.拟合计算圆二色光谱数据发现,与银纳米粒子作用后的血清白蛋白中α-螺旋含量减少,而β-折叠、转角和无规卷曲等结构含量增加,其构象在作用后变得更为松散和伸展.通过时间扫描吸收光谱跟踪研究发现,在血清白蛋白与银纳米粒子相互作用的过程中,银纳米粒子的包覆与聚集、血清白蛋白的构象变化具有双重滞后效应. Interraction and correlation effect of silver nanoparticles and serum albumin were reached by circular dichroism spectrum .Research on surface plasma resonance absorption peak showed that electrostatic forces and hydrophilic force is main force between serum albumin and silver nanoparticles. By fitting conculating data of circular dichroism absorption ,we found that α–helix content reduced ,but structure content such as β–pleated sheet, β–bend and random coil added ,and its conformation became more loose and unfoiding in serum albumin interacted with silver nanoparticle. By tracking time scanning absorbtion spectrum ,we found that package and aggregation of silver nanoparticles , conformation varying of serum albumin have double lag effect during silver nanoparticles interacting with serum albumin.
