摘要
目的研究人间充质干细胞(MesenchymalStemCells,MSCs)的定向分化为成骨细胞的条件及其成骨活性。方法采用标准FicollHypaque技术分离脐血和成人骨髓MSCs,以地塞米松、β磷酸甘油和维生素C为辅剂定向诱导成骨细胞,碱性磷酸酶、骨矿化结节和I型胶原作为成骨细胞鉴定与活性评价的指标。结果两种MSCs的细胞形态和生物学特性无明显差异。定向成骨细胞诱导后,成骨细胞标志性产物碱性磷酸酶、骨矿化结节和I型胶原均得到了稳定的表达,并显著高于未诱导MSCs。结论脐血和成人骨髓MSCs在适当的条件下均可向成骨细胞定向转化。
Objective:To investigate the osteogenic potential of the osteoblasts induced from human umbilical cord blood(HUCB) and adult bone marrow MSCs.Method:The mononuclear fraction was isolated using standard Ficoll Hypaque techniques. Both kinds of cells were purified,proliferated and cultured in the same culture medium.Dexamethasone,ascorbic acid 2 phosphate and β glycerophosphate were used for osteoblasts inducing.Alkaline Phosphatase (ALP) ,type I Collagen and mineralization in vitro were applied to identify the osteoblast and evaluate its activity.Result:Human MSCs could be isolated from HUCB and bone marrow MSCs and these cells had no differences in their origin, morphology,and biological properties. The osteoblasts induced from MSCs had a positive staining for known markers:ALP and mineralization in vitro and also stable expressing of ALP and type I Collagen.Conclusion:MSCs can be induced into osteoblasts in a suitable condition,umbilical cord blood may prove to be a new source for cellular therapeutics for bone repair.
