背景:探讨低密度脂蛋白(lowdensitylipoprotein,LDL)亚组份与人脐静脉内皮细胞(humanumbilicalveinendothelialcells,HUVEC)的血管细胞黏附分子-1(vascularcelladhesionmolecule-1,VCAM-1)表达之间的关系,初步探讨LDL亚组份致动脉粥样硬化(atherosclerosis,AS)作用可能的分子机制。目的:观察不同LDL亚组分对HUVECVCAM-1表达的影响。设计:随机对照的实验研究。地点和对象:本实验在北京市心肺血管疾病研究所动脉硬化研究室进行,体外培养HUVEC。干预:体外培养HUVEL,分别加入25,50,100mg/L的小而密低密度脂蛋白(smalldenselow-densitylipoprotein,sLDL)、大而轻LDL、氧化sLDL和氧化大而轻LDL,共孵育12h和24h。主要观察指标:采用细胞表面ELISA和RT-PCR分别测定VCAM-1蛋白表达及mRNA水平。结果:HUVEC经25,50,100mg/L的sLDL刺激12h后,VCAM-1表达呈浓度依赖性明显增高(0.233±0.036,0.260±0.052,0.365±0.036,F=20.883,P<0.05)。刺激12h后,与大而轻LDL0.231±0.075,oxsLDL0.287±0.034及氧化大而轻LDL0.258±0.043相比,sLDL明显诱导HUVEC的VCAM-1表达(F=17.211,P<0.05)。结论:sLDL显著诱导人脐静脉内皮细胞VCAM-1的表达,可能是sLDL更易引起动脉粥样硬化的机制之一。
BACKGROUND:This study is to explore the relationship between low density lipoprotein(LDL) subclass and vascular cell adhesion molecule 1(VCAM 1) in human umbilical vein endothelial cells(HUVEC),and to primarily investigate the possible molecular mechanism of LDL subclass in atherosclerosis(AS). OBJECTIVE:To observe the effect of LDL subclass on VCAM 1 expression in HUVEC. DESIGN:A randomized controlled experimental study. SETTING and PARTICIPANTS:The experiment was carried out in the Department of Arteriosclerosis,Institute of Cardiopneumoangiogram Disease.HUVEC was cultured in vitro. INTERVENTIONS:HUVECs were cultured in vitro and incubated with 25,50,100 mg/L of small dense low density lipoprotein(sLDL),big particles LDL,oxidized sLDL and oxidized big particles LDL respectively for 12 and 24 hours. MAIN OUTCOME MEASURES:VCAM 1 protein expression and mRNA level was measured with enzyme linked immunosorbent assay(ELISA) and reverse transcription polymerase chain reaction(RT PCR) respectively. RESULTS:VCAM 1 expression was significantly increased with concentration dependence(0.233±0.036,0.260±0.052,0.365±0.036,F=20.883,P< 0.05) after HUVECs were exposed to 25 mg/L,50 mg/L and 100 mg/L of sLDL stimulation for 12 hours.After 12 hour stimulation,as compared with(0.231±0.075) of big particles LDL,(0.287±0.034) of oxidized sLDL and(0.258±0.043) of oxidized big particles LDL, sLDL significantly induced the expression of VCAM 1 in HUVEC(F=17.211,P< 0.05). CONCLUSION:sLDL significantly induces VCAM 1 expression in HUVEC,which might be one of the mechanisms of the induction of sLDL in AS.
