摘要 目的:观察胰高血糖素样肽-1(GLP-1)(7-36)NH2的促胰岛素分泌作用,并进一步探讨GLP-1(7-36)NH2促进胰岛素分泌的相关机制。方法:通过放射免疫分析的方法观察GLP-1(7-36)NH2的促胰岛素效应和GLP-1(7-36)NH2对胞内cAMP浓度的影响。结果:随着GLP-1(7-36)NH2浓度的增加,胰岛素分泌逐渐增加;CAMP类似物8-溴环磷酸腺苷(8-Br-cAMP)增加了胰岛素分泌,GLP-1(7-36)NH2则增加了胞内第二信使cAMP的浓度。结论:GLP-1(7-36)NH2可以促进胰岛素分泌,在一定范围内呈剂量依赖性,其机制与GLP-1(7-36)NH2增加了胞内第二信使cAMP有关。 Objective: To explore the insulin secrection and intracellular cAMP changes stimulated by GLP-1(7-36)NH2. Methods : Determine the amount of insulin secretion stimulated by GLP-1(7-36) NH2 in different concentration and observe intracellular cAMP changes via radio immunoassay analysis. Results: With the rising concentration of GLP-1 (7-36)NH2, insulin secretion increases. Use cAMP analog. 8-Br-cAMP can increase insulin secretion and GLP-1 (7-36) NH2 can increase intracellular cAMP concentration. Conclusion : GLP-1(7-36)NH2 stimulates insulin secretion in a concentration-dependent manner, and it probably relates to intracellular cAMP changes.
