实验性异种角膜移植的免疫学指标测定

摘要 目的通过32磷(32P)标记供体白细胞预处理受体,观察含β射线的32P对异种(鸡→兔)角膜移植免疫学指标的影响。方法建立异种(鸡→兔)角膜移植实验模型,术前2周将32P标记供体白细胞注入兔血管内预处理受体,术后观察新生血管长至植床植片交界处时间,植片透明、半透明和完全混浊时间,同时检测移植家兔外周血白细胞、T淋巴细胞术前术后数量的变化。测定淋巴细胞核内DNA含量及血清抗角膜抗体。结果应用32P标记供体白细胞预处理受体组角膜移植后,植片完全混浊的时间显著延长,但预处理组淋巴细胞数量及淋巴细胞核内DNA含量未见显著降低,而空白对照组则有显著差异,表明小剂量的32P静脉注射是安全的。结论32P标记供体白细胞预处理受体免疫学指标未见显著改变,提示该方法是安全可靠的,为抗角膜移植免疫排斥反应提供了新的手段。 Objective To observe the immunological indexes in xenocorneal transplantation model after pretreatment of receptor by 32P labeled white blood cells. Methods An experimental model of chicken to rabbit xenoco rneal transplantation was established. The 32P labeled white blood cells were intravascular injected to the rabbit to pretreat receptors for two weeks before operation. The time for neovascularization and the clearance of transplanted cornea were observed. The changes of the numbers of white blood cells and T lymphocytes, the DNA content in the lymphocytes and the anti cornea antibodies in sera before and after operation were recorded. Results The time of complete turbidity of transplanted cornea in pretreated group was significantly prolonged (P< 0.01). There were no significant changes in the number of white blood cells and T lymphocytes, the DNA content in the lymphocytes and the anti cornea antibodies in the sera. Conclusion The immunological indexes were not significantly changed after pretreatment of receptor by 32P labeled white blood cell, suggesting that such treatment is safe for cornea transplantation.
