摘要
目的 :探讨水蛭素对体外培养的巩膜成纤维细胞及细胞外基质的影响。方法 :用体外培养巩膜成纤维细胞第 3代 ,将不同浓度的水蛭素 5 0U/ml、4 0U/ml、2 0U/ml、10U/ml、1U/ml分别加入等体积的培养液 2 0 0 μl,每 7孔为一浓度组 ,无药培养液为对照组 ,共 6组 ,培养 4 8小时后用MTT法测定各孔贴壁细胞的吸光值 (A) ,取其上清液用放射免疫法测定Ⅲ型胶原及HA的量。结果 :水蛭素 10U/ml组A值与对照组相比有极显著差异 (P <0 0 1) ,其增生抑制率最高。水蛭素 10U/m组HA、Ⅲ型胶原的浓度明显低于对照组 ,有极显著性差异 (P <0 0 1) ;其余几组中HA的浓度与对照组相比有显著差异 (P <0 0 5 )。结论
Objective:To evaluate the effect of hirudin on the cultured sclera fibroblast and extracellular matrix.Methods:Third generation fibroblast cells were mixed with DMEM and hirudin 50U/ml,40U/ml,20U/ml,10U/ml,1U/ml and the DMEM without hirudin used for comparison.48 hour later,MTT method was used to detect the absorbing light number(A),the liquid was collected,the concentration of Ⅲ collagen and HA was detected with radioimmurroassay. Results:The A value in lou/ml hirudin group was significantly different from one in control group(P<0.01).The arrest rate of cell growth in lou/ml hirudin group was the highest in all hirudin groups.The concentration of Ⅲ collagen and HA in 10U/ml hirudin group was obviously lower than that of other groups,with very significant difference (P<0.01).The differences of HA concentration in other hirudin groups were significant compared with control group(P<0.05).Conclusion:Hirudin has the function of supressing fibroblast and extracellular matrix.It may be useful in the treatment of the ocular proliferative diseases related with fibroblast.
