摘要
目的观察三种生物骨衍生材料移植早期兔外周血T淋巴细胞亚群的变化。方法用复合型完全脱蛋白骨(CFDB)、部分脱蛋白骨(PDPB)、部分脱钙骨(PDCB)三种生物骨衍生材料修复兔桡骨节段性缺损,术后1、2、4周取材,用流式细胞仪检测三种材料移植早期兔外周血T淋巴细胞亚群的变化,并通过常规组织学检测观察4周时三种材料的成骨作用,以新鲜异种骨(FXB)和空白缺损为对照。结果三种材料移植后外周血T淋巴细胞亚群CD4/CD8比值在各个阶段与空白对照组无显著差异(P>0.05),与FXB组有显著性差异(P<0.05)。4周时组织形态观察可见新骨贴附三种材料向缺损方向生长。结论三种材料移植早期未引起严重的免疫排斥反应,且不影响新骨形成,可望用于修复骨缺损。
Objective To analyze the CD4/CD8 ratio changes of subsets of peripheral blood T lymphocyte in rabbits in early period after transplantation of 3 kinds of biotic bone derived materials. Methods Complicated Fully Deproteinized Bone (CFDB), Partially Deproteinized Bone (PDPB), Partially Decalcified Bone (PDCB) were used to repair the segmental defects of radius in rabbits. The flow cytometry was used to record the changes of T lymphocyte subsets of peripheral blood in the rabbits at 1 week, 2 weeks and 4 weeks after operation. The osteogenesis was observed histologically after 3 biotic bone derived materials had been implanted for 4 weeks. Fresh Xenogeneic Bone (FXB) and empty defects were used as the control. Results The CD4/CD8 ratio of peripheral blood T lymphocyte subsets in the biotic bone derived material groups was not significantly different from that in the empty control group (P >0.05), but was significantly different from that in the FXB group(P< 0.05)at all the stages. Histologic observation revealed that new bone formation was adhesive to the material and grew toward defects after 4 weeks. Conclusions The 3 kinds of biotic bone derived materials do not cause serious immunologic rejection in the early period after transplantation. Since new bone formation is not affected, they can be used to repair bone defects.
