人血清补体Cl_r亚基的分离

摘要 人血清中补体第1成分业基的分离及抗血清的制备曾于前文报道,作者在分离了人Cl_s扩亚基制备了抗人Cl_s抗血清的基础上,设计了一个较简便的分离Cl_r亚基的方法,将抗人Cl_s抗血清经DEAE-纤维素层析纯化后,偶联到过碘酸活化的不溶性载体SephadexG-75上,制备成抗Cl_s免疫吸附剂,在Ca^(2+)存在下吸附人血清中的Cl_(rs)成分,再用EDTA螯合掉Ca^(2+),使Cl_r解离下来,从而获得了电泳纯的活性Cl_r亚基成分。 A simple method for isolating the C_(r) subunit of the first component of humen complement is reported.The method allows the recovery of C_(r) on the basis of the specific functional interaction of C_(rs) with anti-C_(s).The antibody of anti-C_(s) was partially purified DEAE-cellulose chromatography.The affinity column was prepared by coupling the anti-C_(s) to periodate activated Sephadex G-75.C_(r) was isolated from small amount of human serum by DEAE-cellulose and affinity chromatography.The final preparation,being hemolytically active and hmogeneous, exhibited molecular weight of 73 kDa as an monomer in SDS-PAGE.
