摘要
血浆样品经0.01mol·L-1盐酸和无水乙醇浸取,在碱性条件下用氯仿萃取,GG-MS-选择离子测量法测定,外标法定量。在选定的色谱条件下,盐酸克仑特罗的检出限为0.05ng,线性范围为0.1-100ng,方法的平均回收率为95.6%,RSD小于6.82%(n=6)。该法毋需衍生化,基质无干扰,简便可靠,适用于生物材料中克仑特罗的快速分析。
The sample of human plasma was treated with 0. 01mol · L -1 hydrochloric acid and alcohol first, and then the clenbuterol in the sample was extracted with chloroform from an sodium hydroxid solution of pH 10 11. The combined extracts of CHC13 solution was treated further as described in the procedure, and a 1μ1 portion from the final sample solution of 100μl in alcohol was taken for the determination by GC/MS with the selective ion measurement method. Under the optimum conditions, linearity was held in the range of 0. 1-100ng of clenbuterol in 1μl of the sample solution, and the detection limit (S/N=3) was found to be 0. 05ng. Recovery test was made by the standard addition method and an average value of 95.6% was obtained, and the RSD s (n=6) obtained were less than 6. 82%. The special features of the proposed method were that no pre-derivatization is necessary, and no interference by the base-material was observed. It was proved that this method is simple and accurate, and is feasible to be used as a rapid routine method for the determination of clenbuterol in human plasma and other biological samples.
