摘要
横纹肌肉瘤是一种高度恶性的软组织肿瘤,化疗是最主要的辅助治疗手段.然而,肿瘤细胞对药物产生多药耐药常常导致治疗失败.本实验以长春新碱为诱导剂,在体外成功建立了耐药细胞系DD/VCR,为进一步研究横纹肌肉瘤的耐药机理制作了模型.耐药前后细胞超微结构发生显著变化,mdr1及mrp基因,P-糖蛋白及多药相关蛋白的表达与横纹肌肉瘤多药耐药的发生有关.
Rhabdomyosarcoma is a malignant soft tissue neoplasm. Chemotherapy constitutes the chief supplementary method in the treatment of rhabdomyosarcoma. However, the appearance of multidrug resistance (MDR) is a major obstacle to successful chemotherapy. In the present study, a multidrug resistant cell line cell line RD/VCR, which showed a typical resistant phenotype to anti-cancer drugs, was induced in vitro by vincristine (VCR). Morphological changes were found in the ultra structure. The expression of mdrl and mrp gene, P-glycoprotein (P-gp) and multidrug resistance associated protein (MRP) were detected by RT-PCR and immunohistochemistry respectively. The mechanism of MDR in rhabdomyosarcoma might be associated with the expression of these genes and proteins.
