摘要
目的 研究缺血再灌注诱导体外培养大鼠心肌细胞凋亡及凋亡相关基因表达。方法 采用体外培养新生大鼠心肌细胞 ,随机分为正常对照组 (Ⅰ组 )、模拟缺血 2h组 (Ⅱ组 )、缺血 2h后再灌注 1h组 (Ⅲ组 )以及持续缺血 3h组 (Ⅳ组 )。TUNEL法检测心肌细胞凋亡 ,荧光显微镜观察心肌细胞凋亡的形态学特征、免疫组织化学法检测Bcl- 2 /Bax基因表达。结果 心肌细胞I/R后 ,TUNEL法检测到阳性凋亡细胞 ,且持续缺血 3h组与再灌注组凋亡指数明显高于缺血 2h组。荧光显微镜观察到典型的细胞凋亡超微结构 ;免疫组织化学检测发现Bcl- 2蛋白表达下调 ,Bax蛋白表达上调。结论 缺血和缺血再灌注均能诱导心肌细胞凋亡 ,且心肌细胞凋亡与Bcl- 2 /Bax基因表达有密切关系。
Objective To study cardiocytes' apopotosis induced by ischemia-reperfusion and its interrelated gene expression. Methods The neonate rat's cardiocytes cultured in vitro were divided into normal control group(1st group),simulating Ischemia-2-hour group (2nd group),reperfusion-1-hour group after ischemia-2-hour (3rd group) and continuing ischemia-3-hour group(4th group) stochastically.Apoptosis were detected by terminal deoxynucleotidy-transferase mediated biotin-dutp nick-end labeling(TUNEL),and its morphology features were observed by fluorescent microscope,Bc1-2/Bax protein expressions were detected by immunohistochemical and image analysis. Results In the 2 nd and 3 rd group,there were masculine apoptosis,and the apoptosis indexes of the 4 th and 3 rd groups were higher than those of the 2 nd group.Typical apoptosis' altrastructure was observed by fluorescent microscope;Bc1-2 protein expression was in down regulation,but Bax protein expression was in the opposite direction. Conclusion Cardiocytes' apoptosis can be induced by both ischemia and ischemia-reperfusion,it has a very close relationship with Bc1-2/Bax protein expressions.
