摘要
目的 本文主要研究 8Gy的60 Co γ辐射对大鼠体内及体外MΦ中NO的产生、iNOS表达、iNOSmRNA转录等的影响。方法 用电子自旋共振 (ESR)技术观察对 8Gy的60 Co γ辐射MΦ中NO产生变化 ,免疫组化法研究MΦ中一氧化氮合酶 (iNOS)表达 ,原位杂交 (ISH)检测研究MΦ中iNOSmRNA的转录。结果 60 Co γ辐射大鼠不仅可使体内MΦ的诱导产生大量减少 ,而且 ,诱导MΦ中NO的产生也相对有所减弱。体外诱导MΦ受辐射后产生的NO有所减少 ,MΦ中iNOS表达减弱 ,iNOSmRNA转录降低 ,但LPS和IFN可在一定程度上扭转辐射的这种效应。结论 8Gy的60 Co γ辐射可引起大鼠体外MΦ中NO的产生、iN OS表达、iNOSmRNA转录等的变化。
Aim The effects of 8 Gy 60 Co-γ irradiation on macrophages (MΦ),including its NO production in vivo and in vitro,expression of iNOS and iNOS mRNA transcription in vitro,have been mainly studied.Methods Electron spin resonance (ESR)- spin trapping was used to detect the effect of NO generation in 8 Gy 60 Co-γ irradiated MΦ in vitro.Immunohistochemistry and In situ hybridization (ISH) were used to observe the expression and transcription of inducible NO synthase (iNOS) in vitro in MΦ respectively.Results The generation in MΦ decreased of radiated rats in vivo,and NO generation in MΦ decreased after 60 Co-γ irradiation in vivo or in vitro.iNOS expression and iNOS mRNA transcription were attenuated after irradiatim,which conuld be reversed by LPS and IFN-γ.Conclusion 8 Gy 60 Co-γ irradiation can change NO generation and iNOS expression in MΦ in vitro.
