摘要
目的 探索西地那非选择性增加股骨头血流量的可能性。方法 a)狗 16只 ,随机分为 A、B两组 ,A组 10只用于经静脉用西地那非实验 ;B组 6只用于经髋关节外周动脉用西地那非实验。b)氢廓清技术用于定量测定股骨头血流量。结果 a) A组 :高剂量组血流量从 (0 .16 89± 0 .0 2 80 ) m L· min- 1 · m L- 1 增加到 (0 .2 0 39± 0 .0 90 7) m L·min- 1 · m L- 1 ,差异不显著 ;中剂量组血流量从 (0 .15 4 1± 0 .0 4 2 1) m L· min- 1 · m L- 1 增加到 (0 .1783± 0 .0 84 7)m L·min- 1 ·m L- 1 ,无显著差异。b) B组 :低、中、高剂量用药后股骨头血流量增加 ,但仅高剂量用药后血流量 (0 .2 2 92± 0 .0 84 7) m L· min- 1· m L- 1与用药前的 (0 .1330± 0 .0 6 6 1) m L· min- 1· m L- 1比较有显著差异 ;血流量与药物剂量间存在回归关系 ,其回归方程为 :Y=4 .184 X+12 .933。结论 西地那非能增加狗股骨头血流量 ,为其治疗股骨头缺血性坏死疾病临床前评估提供了理论依据。
Objective Nitric oxide(NO) plays an important role in mediating blood flow of femoral head. But the time of effect of drug on increasin g NO is very short so far,our study was to investigate the effect of sidenafil on blood flow of femoral head in dogs. Methods a)16 male mongrel dogs were divided into two groups at random. 10 dogs(Group A)were used for intrav enous sildenafil.6 dogs(Group B)were used for three main hip extracapuslar arter ies infusion of sildenafil. b)Hydrogen washout technique was used to measure changes of blood flow of the femoral head before and after administration of sildenafil. Results a)Group A: The average blood flow of femoral head was increased from(0.168 9±0.028 0) mL·min -1 ·mL -1 to(0.203 9±0.09 0 7) mL·min -1 ·mL -1 after administration of higher dose sildenafil,but it was not significant difference;The average blood flow of femoral head was increased from(0.154 1±0.042 1) mL·min -1 ·mL -1 to (0.178 3±0 .084 7) mL·min -1 ·mL -1 after administration of middle dose silden afil,but it was not significant differences. b)Group B: The average blood flow o f femoral head was all incresaed after higher dose, middle dose and lower dose o f sildenafil administration,but it was only significant increase from(0.133 0± 0.066 1) mL·min -1 ·mL -1 to (0.229 2±0.084 7) mL·min -1 ·m L -1 after higher dose of sildenafil administration;The relationship betw een blood flow of femoral head and dose of sildenafil was significant,its regre ssion equation was Y=4.184X+12.933. Conclusion Our study demons trated that sildenafil could increase blood flow of femoral head in dogs,and it was possible to investigate the effect of sidenafil on treatment of avascular n ecrosis femoral head.
