摘要
目的 观察体外培养中亚硒酸钠的浓度变化和培养时间的延长对Bcap 37人乳腺癌细胞增殖及酶活性的影响。 方法 在体外培养Bcap 37人乳腺癌细胞中加入不同浓度的亚硒酸钠 ,常规染色显示其形态和增殖情况及各组Bcap 37人乳腺癌细胞内的酸性磷酸酶、碱性磷酸酶和非特异性酯酶活性。 结果 2 μmol L的亚硒酸钠对肿瘤细胞的生长具有一定的抑制作用 ,而 10 μmol L、5 0 μmol L的亚硒酸钠具有一定的细胞毒性。Bcap 37人乳腺癌细胞内的酸性磷酸酶、碱性磷酸酶、非特异性酯酶活性随着硒浓度的加大 ,培养时间的延长呈下降趋势 ,并且10 μmol L、5 0 μmol L亚硒酸钠培养 2d后 ,3种酶的活性完全消失。 结论 硒对肿瘤细胞的生长和酶的活性都具有抑制作用 ,并且两者之间存在着一致性。
Objective To study the effect of different concentration of Na-2SeO-3 in RPMI-1*!640 medium and different culture time on the growth and enzymes activity of Bacp-37 human mammary tumor cells. Methods In the experiments,cells were cultured and routinely stained to observe the morphology and proliferation of human mammary tumor cells Bacp-37 in vitro and with increasing the concentration of Na-2SeO-3 in RPMI-1*!640 medium,also cytochemical methods were applied to detect the activity of acid phosphatase (AcP),alkaline phosphatase(AlP)and nonspecific esterase(NsE)of human mammary tumor cells Bcap-37. Results 2*!μmol/L Na-2SeO-3 exerts growth inhibitory effects,while 10*!μmol/L and 50*!μmol/L Na-2SeO-3 have cytotoxity effects on the cells.As the Na-2SeO-3 concentration and culture time increased,the three enzymes activity become weaker.Culture in 10*!μmol/L and 50*!μmol/L Na-2SeO-3,two days later,the enzyme activity of tumor cells disappears completely. Conclusion Na-2SeO-3 has inhibitory effects on both cell growth and enzyme activity,and these two effects show consistency.
