摘要
目的 研究鼠源性抗人白细胞介素 2受体单克隆抗体WuTac的功能和药效作用。方法 采用间接免疫荧光法检测WuTac对白细胞介素 2的阻断作用 ;采用同位素法检测WuTac对活化的T细胞增殖的影响 ,并且通过移植物抗宿主的反应 (GVHR)对WuTac体内的药效作用进行评价。结果 WuTac可以明显地抑制人白细胞介素 2的作用 ,并且可以明显地抑制移植物抗宿主的反应。结论 WuTac可以作为一种较为理想的免疫抑制剂 。
Objective To study the function and pharmacodynamics of therapeutical murine anti-human monoclonal antibody WuTac.Methods Study the inhibiting effect of WuTac on IL-2 activity by indirect immunofluorescence, the influence of WuTac on proliferation of activated T cell by isotope method, and the in vivo pharmacodynamics of WuTac by graft versus host reaction (GVHR).Results WuTac significantly inhibited IL-2 activity, T cell proliferation and GVHR.Conclusion WuTac may be used as an ideal immunmodepressant for prevention and treatment of GVHR in bone marrow transplantation and acute rejection reaction in organ transplantation.
