摘要
本文研究了中华绒螯蟹鳃上皮Na+ K+ ATPase的性质 ,并比较了各对鳃丝Na+ K+ ATPase的活性大小。结果表明 :中华绒螯蟹鳃上皮Na+ K+ ATPase的最适温度为 4 0℃ ,温度高于 4 5℃时酶活急剧下降 ;最适 pH为 8.0 ,并且其活力对pH的变化非常敏感 ,当 pH小于7.5或大于 8.2 5时酶活急剧下降 ;最适底物和Na+ ,K+ ,Mg2 + 浓度分别为 1.5 ,75 ,15 ,6mmol/l;反应介质中乌本苷浓度达到 2mmol/l时 ,Na+ K+ ATPase的活力完全被抑制 ;中华绒螯蟹后 3对鳃丝Na+ K+ ATPase的活力极显著高于前
Na +-K +-ATPase has been found in the cell membrane of most aquatic animals. It is the main portion of Na +-K +-pump and closely related to energy metabolizm and transport of ions in and out of cell. There have been many studies on osmotic regulation for the crab Eriocheir sinensis. This paper mainly studies the characteristics of gill Na-K-ATPase of the Eriocheir sinensis. The results show that the optimum reaction temperature of gill Na-K-ATPase in 40℃. The activity of the enzyme drops rapidly when the temperature is higher than 45℃. The optimum reaction pH value is 8.0, and the activity of the enzyme is very sensitive to pH. The zctivity of the enzyme drops rapidly when the pH value is lower than 7.5 or higher than 8.25. The activity of the enzyme is completely restranined when the concentration of the ouabain reaches 2 mmol/l. The optimal concentrations of ATP-Na, Na, K and Mg in the reaction medium are 1.5,75,15 and 6 mmol/l respecitively. The Na-K-ATPase activities of the three posterior pairs of gills are remarkably higher than those of the five anterior pairs.
