摘要
目的 检测SD(Spraquedawley)大鼠吸入烹调油烟 (cookingoilfumes,COF)后肺组织细胞周期和凋亡的变化。方法 以SD大鼠作为实验对象 ,通过动式吸入染毒 ,于染毒后第 11个月末 ,采用流式细胞仪 (flowcytometer,FCM )技术检测肺组织的细胞周期和凋亡。结果 G1期细胞所占百分数随染毒剂量的增加而减少 ,各组间的G1%有显著性差异 (F =5 0 8,P <0 0 5 ) ;S期细胞所占百分数随染毒剂量的增加而增高 ,组间S %有显著性差异 (F =17 0 6 ,P <0 0 1)。各组间肺组织细胞凋亡率有显著性差异 (χ2 =5 1 17,P <0 0 1) ,且存在剂量 -反应关系 (r =0 80 5 ,P <0 0 5 )。结论 在细胞周期中 ,G1期细胞所占百分数随染毒剂量的增加而减少 ,S期细胞所占百分数随染毒剂量的增加而增高 ;
Objective To detect the lung tissue cell cycle and apoptosis of spraque dawley(SD) rats after inhaling cooking oil fumes(COF).Methods SD rats were used to make the chronic toxicity model.Experiment groups were exposed to COF of three kinds of different concentration(6.88±0.31)?(15.06±0.35)?(35.33±1.69)mg/m 3 171 times in 11 months,30min per time.Control group had inhaled clean air.Flow cytometer technique was used to detect lung tissue cell cycle and apoptosis.Results Lung tissue cell apoptosis rate decreased with the rise of COF concentration.There was a significant difference among different dosage groups(χ 2=51.17,P<0.01) with dose-response relationship(r=0.805,P<0.05).G 1 cell percent decreased and S cell percent increased in cell cycle with the in COF concentration rise.There were significant difference among different dosage groups(F=5.08,P<0.05;F=17.06,P<0.01).Conclusion G 1 cell percentage decreased and S cell percentage increased in cell cycle with the rise of COF concentration.The lung tissue cell apoptosis rate decreased with the rising in COF concentration.
