摘要 目的 采用原位杂交技术对脾脏免疫细胞内β 肾上腺素能受体的mRNA进行了检测 ,旨在寻求脾脏免疫细胞表面存在肾上腺素能受体的形态学证据 ,为进一步研究交感神经系统对免疫系统的调节提供证据。方法 对人 2 0例脾脏组织进行连续冰冻切片 ,采用地高辛尾端标记寡核苷酸探针原位杂交技术 (insituhybridiza tion ,ISH)对脾脏组织的免疫细胞内β 肾上腺素能受体mRNA进行了定性检测。用光学显微镜观察了原位杂交结果。结果 脾脏组织内部分免疫细胞存在β 肾上腺素能受体的mRNA ,在脾脏白髓内阳性细胞分布密集 ,红髓内阳性细胞分布极少 ,在白髓内微动脉、边缘窦及淋巴管周围阳性细胞最丰富。结论 在人类脾脏免疫细胞内存在β 肾上腺素能受体的mRNA。 Objective The purpose of the present study was to search for morphologic evidences for β-adrenergic receptor (β-AR) mRNA in cells from spleen tissue,to provide evidences in order to probe into the mechanisms and patterns of the relation between sympathetic system and immune system.Methods The frozen section of 20 cases of human spleen tissue was choosen,the presence of β-adrenergic receptor mRNA in the cells of these spleen tissue was detected by in situ hybridization (ISH).The positive cells was observed using light microscope.Results The distribution of β-AR positive cells in the white pulp were more than those in the red pulp.Much more positive cells were around the vasculature and lymphatic channels.Conclusion β-AR mRNA indeed exist in human immune cells and norepinephrine may have direct effects on immune cells through β-AR on immune cells.β-AR mRNA positive cells are different in spleen red pulp and white pulp.
