摘要 目的:骨髓基质干细胞在体外能定向分化成心肌样细胞。本研究旨在将分离出来的骨髓基质干细胞标记后注射到心壁内,观察其能否分化成心肌样细胞。方法:将骨髓基质干细胞体外培养扩增后用Brd U标记,将标记后的骨髓基质干细胞注射入结扎冠状动脉左前降支远端的大鼠左心室壁内,假手术组仅注射等量的培养液(无细胞)。结果:反复传代可获得大量纤维状的骨髓基质干细胞,Brd U对骨髓基质干细胞的标记率约为41.41%。手术组细胞注射部位心脏组织中部分细胞内可见抗心室肌肌凝蛋白重链单克隆抗体(MHC)、抗Brd U抗体及抗心房利钠肽抗体、抗Brd U抗体双染色阳性颗粒,假手术组仅为单染色阳性颗粒。结论:移植的骨髓基质干细胞能在心脏提供的特殊微环境下分化成心肌样细胞。 Objective: To investigate whether the labeled bone marrow stem cells can differentiated into my-ocardial cells after injected into the mural of heart on the basis of the theory that bone marrow stem cells can differentiated into myocardial cells in vitro. Methods: After ligation of the end of left anterior decending branch , bone marrow stem cells labeled with! Brd U after culture and proliferation in vitro were injected into the mural of left ventricle, the injection of sham-operated groups was cultures instead of cells. Results: Antibody of myosin heavy chain of ventricle and antibody of Brd U, antibody of atrial natriuretic peptide and antibody of Brd U were positive simultaneously in some cells near the site of injection in operation groups, in sham-operated groups only one positive granularity can be observed. Conclusion : Bone marrow stem cells transplanted into heart can differentiated into myocardial cells in the special microenvironments of heart.
