摘要 目的 了解醋氨酚对犬的多器官毒性作用及原因。方法 选用普通杂种犬 ,静脉注射醋氨酚 ,首次剂量 10 0mg kg ,之后每隔 1h 5 0mg kg ,持续 11h ,做血液生理生化和心、肺、肝、肾等组织病理学检测。分为 :对照组、空腹 12h组和空腹 2 4h组。结果 首次注药后 2 4h左右空腹 2 4h组全部死亡 ,72h左右空腹 12h组有 2条死亡。血液生理生化白细胞、谷丙转氨酶 (ALT)、谷草转氨酶 (AST)、乳酸脱氢酶、总胆红素、尿素氮、肌酐升高 ,AST ALT >1,红细胞下降。组织病理心、肺、肝、肾、胃、脾均有淤血 ,左右心室皆见心肌断裂和间质组织水肿。结论 醋氨酚对犬可造成多器官损伤 。 Objective To explore the toxic effects acetaminophen in dogs. Methods Twelve mongrel dogs were divided into 3 groups: control group, 12 h fasting group (Ⅰ), and 24 h fasting group (Ⅱ). Animals in the later two groups were administered intravenously with injection of acetaminophen at the dose of 100 mg/kg and additional doses of 50 mg/kg were given hourly for 11 h thereafter. Physiological and biochemical parameters of the blood were determined and histopathological changes of the heart, lung, liver, and kidney were observed. Results All dogs in 24 h fasting group died at 24 h after the first administration, whereas 2 in 12 h fasting group died at 72 h. The white blood cell count (WBC), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), total bilirubin (TB), blood urea nitrogen (BUN), and creatinine increased (AST/ALT > 1), but the red blood cell count (RBC) decreased. Congestion was found in the heart, lung, liver, kidney, stomach, and spleen. Fragmentation of myocardium and edema of myocardial mesenchyma were also observed. Conclusion Acetaminophen can induce multiple organ injury. The direct cause of heart failure may be resulted from free oxygen derived free radicals.
