摘要
目的初步探讨我国结核分支杆菌的数目可变串联重复位点 (VariableNumberofTandemRepeat ,VNTR)的分布特征及其在基因分型中的应用。方法 选取结核分支杆菌基因组中的 5个VNTR位点 ,采用PCR和琼脂糖凝胶电泳技术 ,并应用BioNumerics(Version3.0 )软件进行处理 ,分析结核分支杆菌DNA中VNTR分布多态性。结果 检测分析的 6 5株结核分支杆菌具有明显的多态性 ,共分成 12个不同的型别 ,其中大部分菌株属于一个型 ,占 6 4 .6 % ,其他菌株呈散在分布。结论 我国结核分支杆菌的VNTR具有明显的多态性。VNTR分型技术具有简便、快速、特异、重复性好等优点 ,可广泛用于结核病的分子流行病学研究。
The distribution,characteristics and the applications in the genotypic study of variable numbers of tandem repeats(VNTR) in Mycobacterium tuberculosis were investigated in the present study.Five loci of VNTR from the genome of M.tuberculosis were selected and analyzed for the polymorphism in distributions by means of PCR,agarose electrophoresis and the BioNumerics (version 3.0) soft ware.The results analyzed in 65 strains of M.tuberculosis isolated from 3 provinces in China showed definite polymorphism in distributions,and 12 different genotypes could be classified in which most of strains belonged to one genotype (64.6%),and the others were scattered in distributions.It is concluded that the VNTR loci in the genome of M tuberculosis isolated in China showed marked polyporphism,and the technique for the study of these loci is a available method for the epidemiological survey on this subject.
