腺垂体同种异体移植实验研究 Ⅱ.新生大鼠腺垂体细胞培养 被引量:3

摘要 腺垂体移植是神经外科学研究中的一个前沿课题,本课题主要利用细胞培养技术,对新生5天以内的大鼠腺垂体细胞进行培养。从实验结果表明,在没有NGF和HRP介入的情况下,以选择培养第6天的腺细胞进行冻存或移植为最佳。 Adenohypophysial transplatation is in the front of the study on neurosurgery. Many researchers in the world have studied it from different standpoint including the determination of graft location,choice of donor age,the source of graft and rejection of transplantaion,and so on.At pressnt,there are two major problem to hinder the development of the study on adenohypophysial transplantaion. One is. the problem of immunogenic rejection;another is the source and preservation of graft. The viewpoint at present is that cultured adenohyphysial cells can be prowided as the source of graft. That can ensure transplantation time and control the number of cells for graft as well. In addition,it is also easy to regulate and control adenohypophysial cell in vitro with NGF and HRP. Moreover,it is advantageous for survival of graft after transplantation,and easy to treat cultured adenohypophysial cells with immunological technique,finally,it improve.the enduranec in the condition of shortage of oxygen and blood. So we have cultured adenohypophysial cells within 5 days rats in order to determine the best time of adenohypophysial cells growth and function in vitro. Results of the experiment showed that adenohypophysial cells kept theis basic morphological characteristic within the 7th days of culture when no NGF and HRP intervened in the medium. Then the morphology of adenohypophysial cells degenerated gradually along with division and growth of fibroblastic cells ,and on the 14th day the have lost their primary characteristic of adenohypophysial cells in morphology. The level of rLH in crlture medin was the highest 42.7±6.9 on the 6th day of cell culture,then decreased gradully to 1.6±0.5 on the 15th day. So our conclusion is that it is the best time on the 6th day of culture of adenohypophysial cells to preserve or transplant.
