摘要 目的 观察联合应用反义 bcl- 2硫代磷酸寡脱氧核苷酸 (bcl- 2 AS- PS- ODN)和白细胞介素 6 (IL - 6 )AS- PS- ODN对小细胞肺癌细胞株 NCI- H4 4 6增殖和活力的影响。 方法 以 bcl- 2 AS- PS- ODN和 IL- 6 AS- PS-ODN单独及联合作用于 NCI- H4 4 6 ,经细胞克隆形成实验、细胞增殖实验观察其对细胞增殖和活力的影响 ,经细胞凋亡线粒体流式检测试剂盒检测细胞凋亡的变化 ,经半定量 RT- PCR检测 IL - 6 m RNA表达水平的变化。 结果 细胞增殖曲线、细胞克隆形成实验及细胞凋亡实验显示联合用药能更强地抑制细胞活力 ,促进细胞凋亡。 bcl- 2 AS-PS- ODN单独作用时 ,IL- 6 m RNA表达上升 74 .3% ,IL- 6 AS- PS- ODN单独作用以及与 bcl- 2 AS- PS- ODN联合作用时 ,IL- 6分别下调 6 7.7%和 5 2 .4 %。 结论 联合用药较单独用药对 NCI- H4 4 Objective To investigate the the combination effect of bcl 2 antisense phosphorothioate oligodeoxynucleotide(AS PS ODN) and interleukin 6(IL 6) AS PS ODN on growth and vitality of the small cell lung cancer cell line NCI H446. Methods Cell clone formation rate and growth curve was used to evaluate growth and vitality of NCI H446 cell line treated with either IL 6 AS PS ODN or bcl 2 AS PS ODN or both of them. Apoptosis was detected by flow cytometry, and semi quantitative reverse transcription PCR was performed to determine the mRNA expression of IL 6. Results Expression of bcl 2 mRNA increased by 74 3% in group of bcl 2 AS PS ODN treatment. Expression of IL 6 mRNA decreased by 67 7% in group of IL 6 AS PS ODN treatment, and decreased by 52 4% while applying with bcl 2 AS PS ODN. Cell clone formation rate and growth curve indicated that combined treatment of IL 6 AS PS ODN and bcl 2 AS PS ODN could more effectively inhibit cell proliferation and induce apoptosis than single drug. Conclusion The conbinative effect of two AS PS ODNs on the NCI H446 cell line was better than single alone.
