摘要
胰岛素抵抗综合征 (insulinresistancesyndrome ,IRS)是以胰岛素抵抗为核心 ,同时并见高血糖、高血压、高血脂或冠心病等多种代谢性心血管疾病的综合征。研究表明 ,IRS的形成与血管平滑肌细胞 (VSMC)凋亡紊乱有关 ,而肌动蛋白是诱导凋亡信号传递的重要因素。养心通脉片能有效提高机体的胰岛素敏感性 ,改善胰岛素抵抗 ,研究表明 ,健康对照组大鼠主动脉的APO和actinr测值均显著低于IRS病理模型组 ,提示IRS形成与细胞凋亡失常及肌动蛋白表达明显增高有关。目的 :研究养心通脉片对胰岛素抵抗综合征 (IRS)大鼠模型主动脉细胞凋亡及肌动蛋白信号传递的影响。方法 :采取“高脂高糖高盐膳食”饲养法复制实验性IRS大鼠模型。大鼠模型随机分为 3组———养心通脉组、二甲双胍组、病理模型组 ,同时设健康对照组。治疗3 0天 ,检测分析各组大鼠主动脉的细胞凋亡和肌动蛋白表达。结果 :4组大鼠比较 ,细胞凋亡率和肌动蛋白表达的平均光密度值都呈现健康对照组→养心通脉组→二甲双胍组→病理模型组递增的趋势 (均p <0 .0 1) ;两治疗组比较 ,养心通脉组的细胞凋亡率和肌动蛋白表达的平均光密度值较之二甲双胍组均显著下调(p<0 .0 1或p<0 .0 5 )。结论 :养心通脉片对主动脉细胞凋亡及肌动蛋白表达的影响明显优于二甲双胍?
Objective:To investigate effects of Yangxin Tongmai Tabellae (YTT) on apoptosis and singnal transmission of actin in aort of rat modelswithinsulin resistance syndrome(IRS).Methods:Using the method of high fat-high glucose-high salt feeding to build the rat models with IRS,the rat models were randomly divided into 3 groups,YTT group,metformin (met) group and pathology group,as healthy controls group,which they were treated 30-days with YTT,met and pure water,respectively.The percentage of apoptosis and expression of actin in the aorta of rat models were measured and analysed compared with healthy controls group.Results:The percentage of apoptosis and average D λnmvalue of actin increased in turns from healthy controls group to YTT group,to Met group,to pathology group(P<0.01).Besides,the percentage of apoptosis and average D λnm value of actin in YTT group were significantly dropper than Met group (P<0.01orP<0.05).Conclusions:Effect of YTT on apoptosis and expression of actin was evidently superiorto Met.
