摘要
目的 研究血管内皮生长因子 (VEGF)基因转染后对Lewis肺癌细胞体外侵袭能力的影响。方法 利用基因重组技术构建了含全长VEGFcDNA的真核双表达载体 ,脂质体法转染Lewis肺癌细胞株。借助Boyden小室膜侵袭培养系统观察转染后Lewis肺癌细胞对基底膜侵袭能力的改变。结果 转染后下室浸润的癌细胞数为 (4 8± 1 .0 ) ,显著高于空载体转染对照组 (2 7± 0 .4 ) (P <0 .0 5 ) ,表明转染pEGFP VEGF表达载体的Lewis细胞系侵袭能力增强。
Objective To explore the effects of vascular endothelial growth factor(VEGF) on invasion potentiality of Lewis lung carcinoma cell line(LLC).Methods A mammalian co expression vector containing VEGF cDNA was constructed and transfected into LLC cells with liposome.Boyden chambers were used for in vitro invasion tests of LLCs transfected with or without VEGF gene.Results After transfection,the abilities of invasion through matrigel in vitro were remarkably increased in VEGF transfected LLCs(58±1.0),comparing with mock transfected LLCs(26±0.4)( P <0.05).Conclusion The increased invasion potentiality of LLC could be induced by VEGF via enhancement of the cell migration.
